W Red Dead Redemption jest w sumie 57 misji głównych czyli takich, które musimy przejść aby zakończyć fabularny wątek gry. Wraz z kolejnymi misjami rozwija się fabuła gry, a gracz zyskuje dostęp do kolejnych misji, obszarów, broni itd. Misję najczęściej zlecają nam ważne dla fabuły postaci oznaczone na mapie inicjałami. Często zdarza się, że w jednym momencie dostępne jest kilka różnych misji, także możemy wybierać w jakiej kolejności je wykonamy.
